چکیده

In this study, the dimensional changes of articular cartilage during cryofixation (immersion-freezing) followed by freeze-substitution and critical-point-drying or tertiary-butyl alcohol drying were compared with conventional fixation techniques. The cryotechnique resulted in 11% shrinkage of the articular cartilage. During conventional chemical fixation, the sample shrank by 28%. No difference in shrinkage could be observed between the two drying methods. Morphologic examination by scanning electron microscopy (SEM) showed no difference between the rabbit tibial plateau cartilage of small excised samples and whole knee joints fixed by the cryotechnique. The collagen structure was unaffected, but the chondrocytes showed formation of segregation artifacts (ice crystal damage) compared to conventional fixation. For the study of articular cartilage with SEM, cryofixation followed by freeze-substitution is superior to conventional fixation methods with respect to specimen shrinkage, but ice crystal artifacts will be introduced into the ultrastructure. It allows immediate preservation of large samples with now damage to collagen structure.

A Comparison of Four Preparation Methods for the Morphological Study of Articular Cartilage for Scanning Electron Microscopy

This study compared 4 fixation methods of rabbit articular cartilage. Tibial plateau samples were prepared either by chemical fixation, with or without microwave enhancement, freezing at ambient pressure followed by freezesubstitution or high-pressure freezing methods. Following the initial preservation, samples were freeze-fractured to expose their internal structure. Microwave enhanced chemic...

Mesenchymal stem cells can survive on the extracellular matrix-derived decellularized bovine articular cartilage scaffold

Objective (s): The scarcity of articular cartilage defect to repair due to absence of blood vessels and tissue engineering is one of the promising approaches for cartilage regeneration. The objective of this study was to prepare an extracellular matrix derived decellularized bovine articular cartilage scaffold and investigate its interactions with seeded rat bone marrow mesenchymal stem cells (...

Costal Versus Articular Chondrocytes in Alginate Three-Dimensional Cultures

Given the difficulties in accessing articular cartilage as a source of chondrocytes to be used in fabricating cartilage constructs, alternative sources are required. The present study examined chondrocytes from costal cartilage for their suitability in cartilage tissue engineering. Chondrocytes isolated from rat knee and rib hyaline cartilage were separately mixed with alginate and placed in a ...

Scanning electron microscopy of normal and rheumatoid articular cartilages.

The fine structure of the surface and superficial layer of the articular cartilage is of special interest for the analysis of the functions and pathological changes of the joint. The arrangement of the collagen fibrils in the surface layer of the articular cartilage has been investigated by the Spaltlinie method (BENNINGHOFF, 1925), by phase contrast microscopy (MACCONAILL, 1951) and by polariz...

Ultrastructural study of upper surface layer in rat mandibular condylar cartilage by quick-freezing method.

The purpose of the present study is to clarify native ultrastructures of upper surface layers of the rat mandibular condylar cartilage in vivo by a quick-freezing method. The mandibular cartilaginous tissues were removed with their articular discs attached without opening the lower joint cavity. The specimens were processed for light microscopy, transmission or scanning electron microscopy. Dee...
